利用组织芯片技术研究丝氨酸/苏氨酸蛋白激酶2在大肠癌中的表达及其临床意义 被引量:1

Expression and clinical significance of protein-serine-threonine kinase 2 protein in human colorectal carcinoma using tissue microarray

摘要 目的探讨大肠癌组织中丝氨酸/苏氨酸蛋白激酶2(AKT2)的表达及其临床意义。方法将50例大肠癌手术标本、10例相应癌旁及14例正常大肠组织制作成组织芯片,同时将17例活检腺瘤标本进行常规切片,应用免疫组化SABC法研究AKT2在大肠癌组织中的表达。结果AKT2在大肠癌组织中的阳性表达率为84%(42/50),明显高于大肠腺瘤的52.9%(9/17)和正常大肠组织的14.3%(2/14),并均有显著性差异(P<0.05、P<0.001)。AKT2的表达与Dukes分期、淋巴结转移有关(P<0.05),而与患者的性别、年龄及组织学分级无关(P>0.05)。结论AKT2在大肠癌组织中表达增高,可能在大肠癌发生、发展、转移中起重要作用。可将AKT2作为临床监控大肠腺瘤癌变及大肠腺癌进展的指标。 Objective To investigate the expression and clinical significance of protein-serine- threonine kinase (AKT2) protein in human colorectal carcinoma. Methods Tissue microarray techniques were made by specimens of 50 cases of colorectal carcinoma, 10 cases of adjacent normal mucosa and 14 cases of normal colorectal mucosa tissue, while 17 cases of adenoma using routine chip method. Expression of AKT2 protein was measured by using immunohistochemical SABC method in them. Results In colorectal carcinoma, the positive expression rate of AKT2 was 84.0% (42/50), while 52.9% (9/17) in adenoma and 14.3%(2/14) in normal tissue, showing the expression of AKT2 protein in cancer tissue was significantly different to that in adenoma and normal tissue, respectively (P 〈0.05, P 〈0.001). The expression of AKT2 protein in colorectal carcinoma was significantly correlated with lymph node metastasis, and dukes stages (P 〈0.05), but not with gender, ages and histological grade (P 〉0.05). Conclusion Overexpression of AKT2 is found in colorectal carcinoma1. AKT2 protein may play an important role in the tumor development and tumor metastasis in colorectal carcinoma. This showed AKT2 may be seen as a clinical monitoring indicators of malignant colorectal adenoma and colorectal carcinoma progressing.
