TGF-α对鼻咽癌CNE2细胞作用的蛋白质组学分析

摘要 目的通过蛋白质组学方法观察转化生长因子α(transforming growth factorα,TGF-α)对鼻咽癌细胞系CNE2蛋白质表达谱的影响,从而解析表皮生长因子受体(epithelial growth factor receptor,EGFR)在鼻咽癌中的作用信号途径。方法以TGF-α作为配体,激活EGFR信号通道,采用无血清细胞培养法,将鼻咽癌CNE2细胞在有或没有TGF-α作用下培养24 h,收集2组细胞总蛋白,通过比较蛋白质组学方法分析蛋白表达谱的变化,质谱鉴定差异蛋白质。结果鉴定了16个差异蛋白,包括EF-Tu precursort、riosephosphate isomerase、nucleoside-diphosphate kinase、annexinA1、hemoglobin beta chain、Heterogeneous nuclear ribonucleoprotein H等,它们广泛参与了信号转导、细胞凋亡与侵袭转移等重要生理过程。结论EGFR可能通过上述差异蛋白所介导的信号转导途径而发挥作用,为鼻咽癌细胞EGFR信号通路的深入研究提供了依据。 Objective By using proteomics, the influence of transforming growth factor α (TGF-α) on protein expression profile of CNE2 was analyzed in order to investigate the epithelial growth factor receptor (EGFR) signal pathway in nasopharyngeal carcinogenesis. Methods Human nasopharyngeal carcinoma cell line CNE2 cells were cultured in serum-free medium, and EGFR signal pathway was activated by TGF-α. CNE2 was cultured with or without TGF-α for 24 h, and the total proteins were harvested. Differential expression profiling was analyzed between the experimental group and the control group, and the differential expression proteins were screened and identified by mass spectrometry. Results Sixteen protein spots were identified successfully as differentially-expressed proteins, including EF-Tu precursor, triosephosphate isomerase, nucleoside-diphosphate kinase, annexin A1, hemoglobin beta chain, heterogeneous nuclear ribonucleoprotein H etc. These proteins were involved in signal pathway, apoptosis, invasion, metastasis and other processes. Conclusion EGFR may be affected by abovementioned proteins. This study provides important basis for the further investigation of EGFR effects and signal pathway in nasopharyngeal carcinogenesis.
