摘要
目的:研究琐琐葡萄多糖(VVP)提取、分离、纯化的方法,探讨其抗氧化能力。方法:建立热水提取、乙醇沉淀、Sevage法除蛋白及透析、纯化VVP的方法,采用苯酚-硫酸法测定其含量,并比较了不同浓度VVP(200、400、600、800、1000μg/ml)和抗坏血酸(Vc50、100、200、400、600、800、1000μg/ml)的还原能力以及不同浓度VVP和VC(均为40、80、160、320、600、800μg/ml)清除二苯代苦味酰基(DPPH)自由基的能力。结果:VVP的还原能力及清除DPPH自由基的能力随其浓度的增加而增大,在800μg/ml时,DPPH自由基清除率达到90%以上,接近VC的清除率。结论:琐琐葡萄多糖具有较好的抗氧化能力。
Objective. To research the method of extract, separation and purification of the polysaccharides from vitis vinifera L. (VVP) and evaluate its antioxidant activity. Methods. Polysaccharides was extracted with water, subsided with 95% ethanol, cleared protein with Sevage assay and then dialyzed and purified. Content of VVP was assayed with phenol-vitriolic colorimetry. Determination and comparing of deoxidiza- tion activities of VVP (200, 400, 600, 800, 1000 μg/ml) and vitamin C (50, 100, 200, 400, 600, 800, 1000 μg/ml) and clearing DPPH free radical activities of VVP and vitamin C (40, 80, 160, 320, 600, 800 μg/ml). Results. With augmentation of concentration, the deoxidization and scavenging DPPH free radical capacity of VVP was increased continuously. VVP(800 μg/ml) could clear 90% DPPH free radical. That was close to Vc. Conclusion. VVP has relatively strong antioxidant activity.
