摘要
目的:分析麦冬类药材总皂苷元含量与抑制中性粒细胞呼吸爆发作用的相关性,考察通过总皂苷元含量预测药材的药效活性。方法:采用HPLC-ELSD法测定麦冬类药材鲁斯可皂苷元及薯蓣皂苷元含量;采用化学发光法测定对中性粒细胞呼吸爆发的作用活性。结果:不同种麦冬类药材在皂苷元组成及含量上均有区别,其总苷元含量从高到低依次为:短葶山麦冬、川麦冬和湖北麦冬、杭麦冬;在活性作用方面:麦冬类药材对中性粒细胞呼吸爆发呈抑制作用,其中短葶山麦冬对中性粒细胞的呼吸爆发具有较强的抑制作用。经分析不同种麦冬类药材的总皂苷元含量与其对中性粒细胞呼吸爆发的抑制作用,总皂苷元含量与抑制中性粒细胞呼吸爆发作用呈相关性。结论:麦冬类药材可根据总皂苷元含量预测对中性粒细胞呼吸爆发的抑制活性,总皂苷元含量是反映药材抗炎活性的一个很好的综合指标,该研究为麦冬类药材的品质评价提供了科学依据。
Aim:To investigate the relationship between the total sapogenin content of various Maidong and its inhibition on neutrophil respiratory burst, and to assess the predictability of the bioactivity based on the content of the total sapogenin. Methods:HPLC-ELSD was employed to determine ruscogenin and diosgenin content of various Maidong. Chemiluminescence method was used to assay the activity of inhibition on neutrophil respiratory burst. Results:There existed differences in sapogenin composition and content among different species. The content of the total sapogenin decreased in the following order: Liriope muscari 〉 Chuanmaidong≈ Liriope spicata var. prolifera 〉 Hangmaidong. On the other hand, various Maidong inhibited neutrophil respiratory burst. L. muscari showed stronger inhibition effect. It was found that there was correlation between the total sapogenin content of various Maidong and their potency of inhibition on neutrophil respiratory burst. The results revealed that the total sapogenin content showed relationship with the activity of inhibition on neutrophil respiratory burst. Conclusion: The bioactive potency could be predicted by the content of the total sapogenin. Total sapogenin content might be used as an indicator for the quality assessment of various Maidong.
