一个葡萄抗逆相关转录因子VvPF1基因的in silico克隆及生物信息学分析

摘要 AP2/EREBP是植物所特有的一类转录因子,广泛参与植物生长、发育以及多种生理生化反应的信号传导。本文利用in silico克隆方法获得葡萄VvPF1基因,并利用生物信息学方法对该基因编码产物从氨基酸组成、理化性质、进化关系、二级及三级结构、功能等方面进行预测和分析。结果表明:VvPF1是一个含有明显AP2结构域,具有潜在核定位能力的ERF类转录因子。此外,VvPF1与CaPF1,JERF1等抗逆相关的转录因子具有较高的同源性,因此推测,VvPF1很可能参与葡萄对生物和非生物胁迫的信号传导。 The AP2/EREBP transcription factors are specifically in plants, which is involved in growth, development and signal transduction of many physiological and biochemical responses. In this paper, a novel transcription factor VvPF1 from Vitis vinifera was cloned by in silico cloning method based on the EST sequences from EST assembly of PlantGDB. Some characteristics of VvPF1 were analyzed and predicted by bioinformatics tools in the following aspects : the composition of ami- no acid sequences, physical and chemical properties, secondary and tertiary structures of protein and functions and so on. Analysis results showed that VvPF1 was a classic ERF transcription factor with conserved AP2 binding domain and a deduced nuclear localization signal (NLS). VvPF1 protein was highly identified with transcription factors involved in resistance, such as CaPFI and JERF1. So we conclude that VvPFI transcription factor might play important roles in signal transduction of bi- otic and abiotic stresses in Vitis vinifera.
