“北京3号”地黄愈伤组织的诱导与分化

以"北京3号"地黄为材料研究不同消毒时间对无菌苗形成的影响及不同激素浓度配比对愈伤组织形成与分化的影响.实验表明:对块根进行消毒的最佳时间为75%乙醇30 s,0.1%升汞10 min.叶片在MS附加6-BA0.5 mg/L、NAA0.3 mg/L培养基上,愈伤组织诱导率可达95%;在MS附加NAA0.1mg/L,6-BA3.0 mg/L的培养基上不定芽的分化率达48.3%.

Tissue culture of Rehmannia glutinosa Libosch, induction of callus were carried out in this work. It can lower the pollution ratio, improve the ratio of germination after the root sterilized with 75% alcohol from 25 to 30s and 0. 1% HgCl2 10 min. Leaf was cultured on MS medium supplemented with 6-BA 0. 5mg/L and NAA 0. 3mg/L, and the callus induction frequency could get to 95%. When the leaves were cultured on MS medium supplemented with 6-BA 3. 0mg/L and NAA O0. 1 mg/L, the adventitious buds arose directly and the differentiation frequency was about 48.3%.
