摘要
目的建立一种特异性强的检测白念珠菌血清抗体方法,协助临床诊断侵袭性念珠菌病。方法应用超声粉碎和ConA-Sepharose4B亲和层析法去除细胞壁甘露糖和糖蛋白后,PAGE电泳显示47000部分是主要的胞浆蛋白抗原,用47000~29000或全菌抗原建立ELISA法测血清抗体。结果全菌抗原,80份正常血清75份阳性,40份患者血清37份阳性;47000~29000蛋白抗原,80份正常血清2份阳性,40份患者血清36份阳性。结论检测血清中抗白念珠菌菌丝相蛋白抗体,有助于诊断侵袭性白念珠菌感染。
Objective To develop a serodiagnostic assay with high specificity to detect antibodies against Candida albicans and differentiate invasive candidosis from colonization of Cndida albicans.Methods Homogenization by sonifier and affinity chromatography on Con A sepharose àB was performed to eliminate the mannan and glycoprotein in cell wall.PAGE showed that the 47 000~29 000antigens or the antigens in the extract from whole cell were used to establish ELISA for detection of antibodies in sera.Results 75 of 80 samples of normal sera and 37 of 40 samples of patients'sera had positive antibodies against the antigens in whole cell.whereas á of 80 and 36 of 40 patients had positive antibodies against 47 000~29 000 antigens.Conclusion The detection of antibodies against mycelial plase protein is helpful for the diagnosis of invasive Candida in fections.
