妊娠期大鼠大量摄入酒精对子代糖代谢的影响及其机制 被引量:2

摘要 目的观察妊娠期大鼠大量摄入酒精对子代出生时及成年期糖代谢的影响,并探讨其机制。方法Wistard孕鼠酒精灌胃4g/(kg.d)至分娩前为酒精组,对照组给予等体积蒸馏水。每组子代鼠在出生1d及13周龄时,测定血糖、血浆胰岛素和抵抗素及脂肪组织抵抗素、瘦素mRNA转录水平。子代鼠13周龄时进行腹腔葡萄糖耐量试验。RT-PCR检测脂肪组织瘦素、抵抗素mRNA转录水平。结果与对照组比较,摄入酒精孕鼠的新生鼠出生时体重明显降低,血糖升高,血浆胰岛素含量降低。13周龄时酒精组血糖与对照组相比差异无显著意义,但血浆胰岛素明显高于对照组;葡萄糖耐量减低,葡萄糖及胰岛素曲线下面积明显增加。血浆抵抗素含量增加,酒精组新生鼠及13周龄子代鼠脂肪组织中抵抗素mRNA转录水平均显著增加,而酒精组新生鼠脂肪组织瘦素mRNA转录水平降低,13周龄子代鼠瘦素mRNA转录水平与对照组相比差异无显著意义。结论孕鼠大量摄入酒精引起子代出生后至成年期糖代谢改变可能与抵抗素增加有关。 Objective To observe the influence of maternal ethanol heavy eomumpfion during gestation on the glyeometabolism of newbem and adult offspring in rats and explore the influencing mechanism. Methods The female Wistard rats during gestation in test group were fed with ethanol at a dosage of 4g/(kg·d) until delivery,and those in control group with distilled water. Determine the blood glucose,plasma insulin and resistin as well as resistin and leptin mRNA levels in fatty tissues of offspring 1 d and 13 weeks after birth respectively. Perform glucose resistance test on offspring aged 13 weeks.Results Compared with those in control group,the bodyweights of newborn rats delivered by female mice in test group were significantly low,the blood glucose level was high,and the plasma insulin level was low.At age of 13 weeks,the blood glucose of offspring of female rats in test group showed no significant difference with that in control group, while plasma insulin levels were significantly higher than that in control group. The glucose resistance levels of adult offspring(aged 13 weeks)delivered by female rats in test group decreased, while the areas under curves of glucose and insulin increased significantly, and plasma resistin level increased, as compared with those in control group. Both the resistin mRNA levels in fatty tissues of newborn and adult offspring of female rats in test group increased significantly, while the leptin mRNA levels decreased. However, the leptin mRNA levels of adult offspring of female mice in test group showed no significant difference with that in control group. Conclusion The change of glycometabolism of newborn and adult offspring caused by maternal ethanol heavy consumption during gestation may be due to the increase of resistin.
