摘要
目的:探讨粒细胞-巨噬细胞集落刺激因子(GM-CSF)对大鼠甲状腺滤泡旁细胞(又称C细胞)的影响。方法:将48只健康SD大鼠随机分为3组,即对照组、小剂量GM-CSF(10μg/kg.d-1)组和大剂量GM-CSF(50μg/kg.d-1)组,每组16只,雌雄各半。腹腔注射,连续7 d,然后用免疫组化S-P法观察大鼠甲状腺C细胞数量。结果:小剂量GM-CSF组C细胞平均数(85.97±20.12)个/HP与对照组(93.58±16.67)个/HP之间无统计学意义(P>0.05),大剂量GM-CSF组C细胞平均数(67.43±13.73)个/HP小于小剂量GM-CSF组(P<0.05);雄性大鼠免疫反应阳性C细胞平均数(88.88±17.16)个/HP大于雌性大鼠(75.78±20.82)个/HP,P<0.05。结论:GM-CSF对大鼠甲状腺免疫反应阳性C细胞的数量具有负性调节作用。
Objective:To investigate the influence of the granulocyte-macrophage colony-stimulating factor (GM-CSF) on rat thyroid parafollicular cells ( C cells). Methods: A total of 48 healthy SD rats were randomly and equally divided into a control group, a low dose group ( GM-CSF 10 μg/kg·d^-1 ) and a high dose group ( GM-CSF 50 μg/kg·d^-1 ). The latter two groups were treated by intraperitoneal administration of GM-CSF at their respective doses for 7 days, and then the thyroid parafollicular cells were stained by immunohishtochemical streptavidin- peroxidase (S-P) technique and their number counted. Results : The number of C cells was(67.43± 13.73 ) in the high dose group, significantly smaller than that of the low dose group(85.97 ± 20.12)(P 〈 0. 05), which was yet smaller than that of the control (93.58 ±16.67). And a larger number of C cells was observed in the thyroid tissues of the male rats ( 88.88± 17.16 ) than in the females (75.78 ±20.82 ). Conclusion : GM-CSF negatively regulates the number of thyroid immunostaining positive C cells in rats.
