摘要 目的探讨牛膝多糖硫酸酯(S-AbPS)和磷酸酯(P-AbPS)衍生物对人肺癌A549细胞的影响及其作用机制。方法用四甲基偶氮唑盐(MTT)法检测牛膝多糖、S-AbPS和P-AbPS对A549细胞增殖的抑制作用;流式细胞仪观察S-AbPS和P-AbPS处理A549细胞后细胞凋亡的改变。结果S-AbPS和P-AbPS对A549细胞的增殖有抑制作用,且存在时间和剂量相关性。S-AbPS和P-AbPS的48 h抑瘤活性均较同浓度的牛膝多糖强。流式细胞仪分析显示,S-AbPS和P-AbPS均可不同程度地诱导细胞凋亡。结论S-AbPS和P-AbPS较牛膝多糖能更有效地抑制人肺癌A549细胞的增殖,其机制可能是通过诱导A549细胞凋亡而实现。 Purpose To study the effect and mechanism of the sulfated derivative (S-AbPS) and the phosphorylated derivative (P-AbPS) of Achyranthes bidentata polysaccharides (AbPS) on human lung cancer cell A549. Methods MTT assay was used to determine the proliferation inhibition by AbPS and its two derivatives on A549. The influence of S-AbPS and P-AbPS on the cell apoptosis was observed with flow cytometry. Resalts The sulfated and phosphorylated derivative of AbPS could inhibit the growth of A549 cell with a dose-effect and time-effect relationship. The cell growth inhibition effect S-AbPS and P-AbPS on human lung cancer. was more strong than that of AbPS with the same dose in 48 h. According to the analyses of flow cytometry, both S-AbPS and P-AbPS could induce A549 cell apoptosis with different levels. Conclusion Both S-AbPS and P-AbPS can inhibit proliferation of cell A549 more effectively than AbPS, and the mechanism may be re- lated to its inducing of apoptosis.
