摘要
应用地高辛标记反意生长抑素cRNA探针原位杂交技术,研究5日龄AA肉鸡生长抑素mRNA阳性神经元胞体在下丘脑内的分布。结果表明:生长抑素mRNA阳性神经元主要分布在下丘脑外侧核吻侧部、后内侧核和后核吻侧部以及乳头体外侧核。生长抑素mRNA阳性神经元胞体为圆形、梭形和椭圆形,直径7~20μm。胞质内均有强度不等的黑色或蓝黑色杂交反应物。生长抑素mRNA神经元胞体在下丘脑各核区的分布数目存在明显差异,其中下丘脑外侧核内的阳性胞体密度高于其它核区,提示下丘脑外侧核的生长抑素神经元的生理功能很活跃。有些核区显示的生长抑素mRNA阳性神经元有突起,说明这些神经元的树突或轴突可能可以储存生长抑素mRNA。
Somatostatin (SS) mRNA positive neurons in the chicken hypothalamus were demonstrated by in situ hybridization histochemistry with digoxigenin labelled antisense SS cRNA probe. The results showed that SS mRNA positive neurons were distributed in the rostral parts of nucleus lateralis hypothalami (LH), nucleus medialis hypothalami caudalis, the rostral parts of nucleus hypothalami caudalis and the nucleus mamilaris lateralis. The SS mRNA positive cell bodys were fusiform or elliptical in shape and 7 20 μm in size. There were different densities of hybridized reaction products in perikaryon. The number of SS mRNA positive neurons in different nucleus varied. The density of positive neurons was higher in the LH than in the other nucleus. These results showed that the physiological function in the LH was more active. The processes were also seen in many SS mRNA positive neurons, suggesting that the dendrite or axon of neuron may store SS mRNA.
