盐胁迫对大豆发育子叶愈伤组织的生化影响 被引量:19

摘要 在含不同浓度NaCl的MS培养基上,继代5次培养大豆发育子叶的愈伤组织,分析了不同盐浓度培养的大豆愈伤组织的蛋白质、氨基酸和可溶性糖的含量。经测定表明,盐胁迫下,大豆愈伤组织的蛋白质,总氨基酸含量都比对照高,可溶性糖含量比对照低。NaCl浓度在0~0.1%范围内,随NaCl浓度的增加,蛋白质与总氨基酸的含量逐渐增加,当NaCl浓度增至0.15%时,蛋白质和总氨基酸含量又降低。在含NaCl0.07%培养基中,大豆愈伤组织的脯氨酸含量最高,为对照的2.72倍。盐胁迫下、脯氨酸、丙氨酸、精氨酸和组氨酸的含量变化与总氨基酸含量变化不一致。 The young cotyledon soybean callus tissues were cultured in the different concentration NaCl medium. For five generations, the contents of protein, amino and soluble sugar of soybean callus tissue were measured. The result showed that contents of protein and amino of callus tissue under salt stress were higher than those of control. Soluble sugar content was lower than that of control. Except in 0.15% NaCl, the contents of protein and amino were increased with increase of NaCl concentration. The accumulation of proline in 0.07% NaCl medium was 2.72 times higher than in control. The changes of proline, alanine, arginine and histidine were different from the change of total amino under salt stress.
