单克隆抗体 M26~32 腹水有效免疫球蛋白的亚类及等电点的分析 被引量:3

摘要 单克隆抗体杂交瘤细胞株M26~32的高滴度腹水,用MonoQ阴离子交换树脂层析柱进行纯化,并用培养的恶性疟原虫和食蟹猴疟原虫抗原片对纯化后的各管作免疫荧光测定(IFA)。对纯化后单抗的有效万分进行Ig亚类检测和等电聚焦电泳分析,结果证实M26~32单抗的有效成分为IgM,等电点(PI)为4.7~4.85。 Mouse ascites of McAb M26-32 were Purified through Mono Q ion exchange column chromatography and all of the fractions were detected with both P. falciparum and P. cynomolgi slides by IFA test. Determination of McAb isotype and polyacrylamide gel electrophoresis were performed for the positive fractions of McAb M26-32. The results showed that the isotype of effective composition of McAb M26-32 was IgM and the isoelectric point was in the range from pH4.7 to 4.85.
