防龋剂氟化物对牙髓细胞增殖及碱性磷酸酶活性的影响

摘要 用四氮唑溴盐(MTT)染色法和以对硝基苯磷酸脂(PNPP)为底物检测不同浓度NaF对大鼠牙髓细胞的影响。实验结果显示10ppmF-开始对牙髓细胞增殖产生抑制(P<0.01);50ppm影响最大,细胞形态发生改变,圆缩死亡细胞明显增多。F-抑制细胞碱性磷酸酶(ALP)活性,浓度增高,抑制增强。提示一定浓度F-对牙髓细胞某些功能活动具有抑制作用。 Wistar rat dental pulp cells were cultured in fluoride mediums(final concentration 0,5,10,25ppm,respectively)in order to investigate the biological effect of fluoride ion on pulp cell proliferation and alkaline phosphatase(ALP) activity.The results showed that NaF started to inhibit cell growth at 10ppm and the inhibition was most affective at 50ppm when alteration of cellular shape was noticeable,deadly cells were increased.Moreover,NaF concentration dependently in hihibited the ALP activity of the cells.The results suggested that fluoride might inbibit some function of pulp cells when a certain concentration was attained.Thus,fluoride concentration which was directly used to dental pulp should be controlled so as not to damage dentalpulp.
