芎归滴丸对小鼠急性脑梗死的预防作用及其机制分析 被引量:5

摘要 目的观察芎归滴丸对小鼠急性脑梗死的预防作用并探索其可能机制。方法使用光化学诱导小鼠急性脑梗死模型,观察1、10、100mg·kg-1芎归滴丸灌胃14d预处理对急性脑梗死的影响;培养新生大鼠海马神经元,用膜片钳全细胞记录诱发电压门控钙电流,观察芎归滴丸对钙电流的影响。结果经芎归滴丸预处理小鼠的急性脑梗死体积减小,并有剂量依赖性;芎归滴丸细胞外给药能抑制海马神经元电压门控性钙电流。结论芎归滴丸具有一定预防急性脑梗死的作用,其机制可能与抑制神经元电压门控性钙离子通道有关。 Aim To determine the mechanism of a new drug Xionggui Pills (XGP) against cerebral infarction. Methods A acute cerebral infarction model of mice was employed to investigate the effect of pretreatment during the 14 days per os by 1,10,100mg·kg^-1 XGP on the volume of infarction. The whole-cell recording patch clamp technique was used to record the voltage-gated calcium current in cultured hippocampal neurons of rats so as to investigate the current ampli- tude treated by 10^-6 ,10^-5 ,10^-4g·ml^-1 XGP. Results The volume of cerebral infarction pretreated by XGP decreased significantly with the increase of XGP administration, so did the calcium current amplitude. Conclusion It was suggested that XGP pretreatment alleviated cerebral infarction and XGP was a calcium channel blockade.
