摘要
目的:观察内毒素损伤内皮细胞过程中一氧化氮(NO)的变化及其意义。方法:选择体外培养的人脐静脉内皮细胞(HUVEC)为研究对象,分别用内毒素及硝基左旋精氨酸(LNNA)对之进行处理,并检测其上清液中NO-2/NO-3丙二醛(MDA)含量及乳酸脱氢酶(LDH)活性变化。结果:LNNA可使内皮细胞NO合成降低,MDA含量和LDH活性下降,而LPS组上述各项指标则明显上升。结论:内毒素可导致内皮细胞,NO增多并与氧自由基相互作用,从而引发内皮细胞损伤。
Objective: The changes of nitric oxide (NO) during cellular damage by endotoxin and its significance were studied. Methods: After the cultured human umbilical vein endothelial cells (HUVECs) were handled with endotoxin and LNNA respectively, the contents of NO-2/NO-3 and MDA and the changes of LDH activity in its supernatant were determined. Results: LNNA could inhibit the synthesis of NO and decrease the content of MDA and LDH activity. Conclusion: The overproduction of NO induced by endotoxin and the interaction between NO and oxygen free radicals might account for the damage of endothelial cells.
