摘要
目的;丝裂素活化蛋白激酶(MAPK)与大鼠胸主动脉内皮剥脱术后平滑肌细胞(SMC)增殖的关系。方法:将剥脱术后的Wistar大鼠随机分为两组(n=6),分别于术后7天和14天时处死取材,其血管条分别做3H-TdR参入和MAPK活性测定,并与假手术组相比较。结果:剥脱术后7天和 14天与假手术组相比,3H-TdR参入分别是后者的 2.6倍和 2. 0倍(P<0. 01),MAPK活性分别为 7.6倍和 2. 4倍(P<0.01)。结论:大鼠胸主动脉球囊内皮剥脱术后血管SMC发生增殖,同时MAPK活性明显升高,说明MAPK与内皮损伤后的血管壁SMC增殖有关。
Objective: To study the relationship between the proliferation of vascular smooth muscle cells (VSMC) and activation of mitogen-activated protein kinase (MAPK) after endothelial denudation with balloon. Methods:Wistar rats after angioplasty were randomized into two groups (n= 6) and the animals were killed on the 7th day and the 14th day,respectively. The activity of MAPK and the incorporation rate of 3H-TdR of injured vascular tissue were measured and compared with those of the sham group.Results:The activities of vascular MAPK on the 7th day and the 14th day after angioplasty were 7. 6 and 2. 4 times higher than those of the sham group (P<0. 01),respectively,and the incorporation rate of 3H TdR of these two groups was 2. 6 and 2. 0 times higher than that of the shatn group(P<0.01),respectively. Conclusion: The proliferation of VSMC occures after endothelial denudation and the vascular MAPK is significantly activated and increased after endothelial denudation. The results indicate that activation of MAPK is involved in the VSMC proliferation after angioplasty.
