葡萄糖调节蛋白75基因对H_2O_2诱导的C6胶质瘤细胞氧化应激性损伤有保护作用 被引量:1

摘要 目的:研究葡萄糖调节蛋白75(grp75)基因对H2O2诱导的C6胶质瘤细胞氧化应激性损伤的保护作用。方法:以grp75基因瞬时转染C6胶质瘤细胞,以H2O2作为氧化应激刺激物,建立细胞损伤模型,运用MTT、LDH、苔盼兰拒染、细胞周期流式细胞术(FCM)和凋亡小体方法观察grp75对H2O2引起的C6细胞氧化应激性损伤的影响。结果:grp75转染组C6细胞在H2O2(0.1mmol/L,24h)刺激后MTT增加、LDH活性降低,细胞凋亡明显抑制,与空载体转染组相比有显著差异。结论:grp75基因对H2O2引起的C6细胞氧化应激性损伤有保护作用。 Objective:To investigate the protective effects of grp75 gene on hydrogen-peroxide (H2O2)-induced cell damage in rat glioma C6 cells. Methods-Cultured C6 glioma cells were transfected with grp75. Assays of MTT,LDH,FCM analysis and hoechst staining were used to investigate the protective effect of grp75 on H2O2 stimulated C6 cells. Results:Transfection of C6 glioma cells with grp75 resulted in the increased cell viability and decreased apoptosis ratio as compared to the control. Conclusion:These data indicated that grp75 might inhibit H2O2-mediated cytotoxic effect on C6 cells.
