海人酸致大鼠海马及脑脊液内氨基酸的变化疒间 被引量:1

摘要 目的:研究海人酸(KA)致疒间大鼠模型海马及脑脊液内谷氨酸(Glu)和天冬氨酸(Asp)的变化,方法:SD大鼠随机分成二组,KA组:KA(2μg/kg)海马杏仁核注射后分为6h、1、3、7天组,每组8只;9g/L盐水对照组(n=8)。采用高效液相色谱(HPLC)法测定大鼠海马组织及脑脊液中兴奋性氨基酸(Glu和Asp)水平变化。结果:海马中Glu、Asp的水平在观察的24h内随时间推移呈逐步增长趋势,但与对照组相比统计学上无显著性差异,3天后仅Glu较对照组明显上升。脑脊液中Glu、Asp的水平在致疒间后6h便较对照组有显著差异,且7天内保持上升趋势。结论:癫疒间大鼠脑脊液中兴奋性氨基酸水平变化较海马组织内早。其中Glu更具有代表性。 Objective:To study the changes of glutamate (Glu) and aspartate (Asp) in hippocampus formation and CSF on KA induced rats. Methods:Sprague-Dawley rats (n = 40) were divided into 2 groups at random:the KA group (intracerebroventricular injection (icy) of KA ,2 μg/kg) and the control group (icy of NS). KA group were divided into 4 groups at 6 h, 1d,3d and 7d,eight in each group. HPLC was used to assess the concentration of Glu and Asp in hippocampus formation and cerebrospinal fluid. Results.In the hippocampus,the contents of Glu and Asp increased continuously ld after seizure,but had no significance compared with the control. Three days later,only Glu became significantly different from the control. However,the contents of Glu and Asp in the CSF were significantly different from the control 6 hours after seizure. Conclusion:The contents of excite amino acid(especially Glu)in CSF increased immediately after KA injection,which changed earlier than those in hippocampus formation.
