摘要
为获得满足工业化生产要求的D-核糖发酵菌株,从腺嘌呤营养缺陷型(aDe-)枯草杆菌Bacillus subtilisXI-335出发,通过目的基因内部缺失和插入失活以及DNA重组技术,获得了转酮醇酶基因(Transketolase,tkt)失活的枯草杆菌菌株XH-56,在适宜的条件下,该菌株能够积累D-核糖,浓度为65.2mg/ml,为出发菌株的36.2倍,已经达到发酵生产用菌的水平。
To obtain a strain which met the requirement of the industrial production of D-ribose fermentation, an adenine auxotroph (ade-), Bacillus subtilis XI-335, was modified by internal gene deletion and insertion inactivation of DNA, then a recombinant, named strain XH-56, was obtained in which transketolase gene was inactivated. Under a suitable condition, the strain XH-56 was able to accumulate D-ribose of 65.2 mg/ml, and the concentration reached the level of mass production fermentation.
