摘要
文章表达、纯化了重组人源肝星状细胞激活相关蛋白,研究了其抗氧化的生物活性和对次氮基三乙酸铁造成的大鼠肝星状细胞氧化应激的抑制作用。结果显示重组人源肝星状细胞激活相关蛋白具有抗氧化的生物活性,活性为(80.0±3.0)U/mg;可抑制次氮基三乙酸铁对肝星状细胞造成的氧化应激,具体表现为可以降低丙二醛、羟脯氨酸的含量,提高超氧化物歧化酶、谷胱甘肽过氧化物酶的活力,并且抑制肝星状细胞的增殖。重组人源肝星状细胞激活相关蛋白对肝星状细胞氧化应激的抑制作用具有剂量依赖关系,浓度大于或等于10μg/mg,即有明显的作用。
To study the bioactivity of recombinant human stellate cell activation-associated protein(rh- STAP) in vitro, expressing and purifying the rhSTAP, studying the antioxidant activity of rhSTAP, and studying the inhibitory action of rhSTAP on HSC oxidative stress induced by Fe-NTA. The rhSTAP has the antioxidant capacity, which is (80.0±3.0)U/rag; the rhSTAP can obviously relieve oxidative stress caused by Fe-NTA, lower the level of MDA and Hyp, raise the activity of SOD and GSH-Px, and inhibit HSC's proliferation. The experiment result indicates that the antioxygen biology activity of rh- STAP can obviously relieve the oxidative damage caused by Fe-NTA, and by relieving oxidative damage, inhibit the oxidative stress reaction of HSC, and by this way inhibit the activation of HSC. It is embodied as improving the organism's anti-oxidative damage system, lessening the lipid peroxidation, decreasing the synthesis of collagen protein, and inhibiting the proliferation of HSC. The protection effect of rhSTAP has a dose-dependent relationship. When the concentration is or is more than 10 μg/mg, the obvious effect comes out.
