摘要
为探索浮膜培养法生产卡提素新工艺,用液体苏通作培养基培养卡介苗菌,收集菌体,按卡提素提取工艺提取。同时,以传统的固体培养法(苏通马铃薯培养基)作参照,比较两种方法所得卡提素的提取率和卡提素的理化指标。结果显示:浮膜培养法所得卡提素的提取率为1.61%,比固体培养法的提取率3.08%低,理化指标符合精制卡介菌多糖核酸标准规定,其中核酸含量明显高于固体培养法的含量。而且,浮膜培养法手工操作量小,质量更容易控制,是一套适合卡提素工业化生产的新工艺。
To explore the velum cultivation method, a new technique to produce BCG-PSN by cultivating BCG in Sauton's medium and then by extracting it following the normal BCG extraction procedures. A comparative study on the extraction percentage and the physicochemical indexes of the solid cultivation method(Sauton's potato medium) and the velum cultivation method was made. Resuets indicated that BCG-PSN extraction percentage of the film cultivation method was 1.61 percent, lower than that of the solid cultivation method by 3.08 %. However the nucleic acid of the new method obviously exceeded the old in content and its physicochemical indexes conformed to purified BCG-PSN standards as well. The author concludes that the velum cultivation method, characterized by less manual work and easier quality control, is more suitable for mass production.
