摘要
单细胞凝胶电泳技术是一种简便、快速、灵敏的检测真核细胞DNA损伤与修复的方法。其基本原理是将单个细胞包理于琼脂糖凝胶中进行裂解、电泳、荧光染色和显微镜观察,细胞DNA损伤产生的断链或碎片在电泳中迁移形成典型的"慧星"图像,根据迁移长度和荧光强度即可定量分析DNA损伤的程度。该文总结了该实验技术的基本原理、检测方法及其在临床兽医学中的应用前景。
The single cell gel electrophoresis is a simple,sensitive and rapid technique for measuring DNA damage and repair in individual cell.The basic principle: single cells,being embeded in agar gel,were decomposed,electrophoresed,dyed by fluorescence and observed with the microscope.The broken chains or pieces induced by DNA damage were moved and formed the typical "comet" picture in the electrophoresis.Then DNA damage was quantitatively analyzed according to its' moving length and fluorescence intensity.The basic principle,development,detection method,advantage and application prospect of the technique in clinical veterinary medicine were reviewed.
