摘要
目的:研究杏仁基底外侧核(BLA)及杏仁中央核(CeA)对大鼠摄食行为及体质量调控的影响.方法:采用电解毁损方法对成年雌性大鼠分别行BLA双侧毁损或CeA双侧毁损,对照组行假毁损手术,术后连续10d记录摄食量及体质量变化,实验结束后灌流动物,取脑组织,切片染色,对毁损部位进行定位.结果:对照组大鼠施行假手术后体质量略有下降,之后呈现缓慢上升趋势,动物摄食量呈增大趋势,日平均摄食量1~3d为(17.9±0.8)g,4~10d为(22.0±0.3)g.毁损BLA组术后体质量呈明显上升趋势,平均增长(24.4±1.4)g(P<0.01);日平均摄食量也较对照组显著增多,1~3d为(20.1±0.7)g,4~10d已增大到(24.6±0.4)g(P<0.01).毁损CeA组术后体质量呈明显下降趋势,体质量减轻程度明显大于对照组(P<0.01);大鼠的日平均摄食量也较对照组明显减少,1~3d为(15.6±1.0)g,4~10d为(19.8±0.4)g(P<0.01).结论:CeA可引起食欲,促进摄食并增加体质量;而BLA的作用与其相反,可降低食欲,抑制摄食并控制体质量增长.
AIM : To investigate the roles of basolateral amygdala (BLA) and central amygdala (CeA) in feeding behavior and body weight control of rats. METHODS: Electrolytic lesions were made bilaterally at BLA or CeA respectively in the mature female rats, while sham lesions were made in the rats of the control group. Food intake and body weight were measured daily for 10 d after the surgery. At the end of the experiments, rats were transcardially perfused, and the brain tissues were fixed, frozen, cut into 40-μm serial sections and stained with neutral red. The sites of the lesions were reconstructed. RESULTS: The body weight of the rats in the control group decreased slightly after the sham-lesion surgery, then showed a slow increasing trend; the food consumption also presented a rising trend with the mean food consumption at 1-3 d as (17.9 ±0.8) g, and at 4-10 d as (22.0 ± 0.3 ) g. The body weight of the rats in BLA-lesioned group showed an obvious increasing trend after the surgery. The mean weight change was (24.4±1.4) g (P〈0.01). The food intake was more than that of the rats in the control group. The mean food consumption at 1-3 d was (20.1 ± 0.7 ) g and ( 24.6 ± 0.4) g ( P 〈 0.01 ) at 4-10 d. The body weight of the rats in CeA-lesioned group showed a slow decreasing trend after the surgery. The weight loss was greater than that in sham-lesioned rats ( P 〈 0.01 ). The food intake was less than that of the rats in the control group. The mean food consumption at 1-3 d was ( 15.6 ± 1.0) g, and (19.8 ±0.4) g (P〈0.01) at 4-10 d. CONCLUSION: CeA stimulates feeding behavior and increases body weight, while BLA inhibits feeding behavior and decreases body weight.
