左归丸对痴呆鼠氧化反应及细胞凋亡的影响 被引量:8

摘要 目的探讨左归丸对AD鼠氧化反应及细胞凋亡的影响。方法模型组以β淀粉样蛋白(Aβ)注射海马制备AD动物模型,各治疗组分别在造模的同时灌胃左归丸和抗脑衰胶囊、脑复康。采用生化法检测各组大鼠脑匀浆CAT,MAO,免疫组化方法观察各组大鼠海马神经元TNF-α蛋白的变化,运用RT-PCR测定各组海马caspase3基因的表达。结果灌服后左归丸治疗组均能明显提高AD大鼠脑组织CAT活性、抑制MAO活性;中药治疗组能显著下降海马区域TNF-α含量和下调caspase-3 mRNA的表达。结论左归丸对痴呆鼠海马神经元细胞凋亡有抑制作用,其作用机理可能是通过提高CAT等抗氧化酶作用和抑制MAO活性,下降海马区域TNF-α蛋白含量和下调caspase-3 mRNA的表达。 Objective To investigate the effects of Zuoguiwan on antioxidation and Apoptosis of Alzheimer's disease(AD) model rats. Methods The hippocampus injected β - amyloid(Aβ) to make AD models. Every group was fed with Naofukang, Kangnaoshuai and Zuoguiwan. The contents of CAT and MAO in the brain were measured by the method of biochemistry and molecular biology. The expression TNF - α protein of hippocampus neurons was observed by electron lens using immunohistochemistry method. The expression of caspase - 3 gene was measured by the methods of RT - PCR and agarose gel electrophoresis. Results - Zuoguiwan could reduce the activity of MAO and improve the activity of CAT in the brain. Zuoguiwan could reduce the expression of caspase - 3 gene and TNF - α protein of Hippocampal neurons. Conclusion Zuoguiwan has repressive effects on apoptosis of Hippocampal neurons in AD model rats. The effective mechanism may associate that Zuoguiwan reduces the activity of MAO and improves the activity of CAT in the brain, reduces the expression of caspase - 3 gene and TNF - α protein of Hippocampal neurons.
