土鳖虫含药血清与化疗药物联合对人肝癌细胞增殖的影响 被引量:2

摘要 目的:观察土鳖虫含药血清作用前后肝癌HepG-2细胞化疗药物相关基因表达水平的改变,探讨其可能的分子机制。方法:以肝癌HepG-2细胞为研究对象,使用实时荧光定量RT-PCR方法检测土鳖虫含药血清作用前后肝癌HepG-2细胞化疗药物相关基因TS、ERCC1表达水平的变化。采用MTT法检测10%土鳖虫含药血清与5-FU、DDP单独、联合用药对肝癌HepG-2细胞增殖的抑制作用。结果:实时荧光定量RT-PCR结果显示,10%土鳖虫含药血清对TS基因表达有下调作用,对ERCC1基因表达有上调作用。药物敏感实验中,在单用土鳖虫含药血清对肝癌HepG-2细胞还未产生抑制作用时,联合5-FU或DDP用药抑制率即高达69.315%或74.775%,显示土鳖虫含药血清与5-FU或DDP联合用药有明显的协同作用。结论:土鳖虫乳剂作为一种新型抗肿瘤中药制剂,经过动物体内代谢后所得血清与常用化疗药物5-FU、DDP有明显的协同作用,可能与药物相关重要基因表达的变化有关,但与DDP协同作用的机理有待进一步研究。 Objective: To inestigate the influence of the eupolyphaga cream contained serum on chemotherapeutic agents associated genes in vitro. Methods: The human hepatoma HepG-2 cell lines was used in the study. The influence of the serum on chemotherapeutic agents associated genes (TS and ERCC1 ) was measured with real time quantitative PCR (RT-PCR). MTT assay was used to authenticate the result of the gene test. Results: The anticancer drug associated gene such as TS was down regulated, while ERCC1 was up regulated. The treatment of HepG-2 cell with the serum combination with 5-FU or DDP could significantly increase the antineoplastic activity in vitro. Conclusion: Eupolyphaga cream as a new poweful antineoplastic herb praeparatum promotes the development of traditional Chinese medicine anticancer ressarch.
