再生障碍性贫血和真性红细胞增多症患者TCRζ链基因表达情况

摘要 目的:分析再生障碍性贫血(AA)和真性红细胞增多症(PV)患者外周血T细胞中TCRζ链基因表达水平,从而了解患者T细胞活化过程的相关变化情况。方法:利用SYBR GreenⅠ荧光定量PCR,相对定量检测25例AA患者、12例PV患者及30例正常人的外周血单个核细胞(PBMCs)的TCRζ链基因表达情况,β2微球蛋白基因(β2M)为内参照,根据相对定量公式:2-△△Ct计算患者与正常人TCRζ链基因表达差异倍数。结果:与正常人相比,TCRζ链基因表达水平有40%AA(10例)和41.7%PV(5例)患者呈表达上调,其它的(60%AA和58.3%PV患者)则呈表达下调情况,TCRζ链基因表达水平的变化与患者年龄无相关性。结论:AA和PV患者可能存在T细胞活化相关的信号通路紊乱的情况,TCRζ链基因表达模式改变的意义仍需进一步研究。 Aim:To investigate the expression level of TCR ξ chain gene in T cells from patients with aplastic anemia (AA) and polycythemia vera (PV), thereby to estimate the feature of T ceils activation status. Methods: Real-time PCR with SYBR Green I technique was used for detecting the expression level of TCR ξ chain gene in peripheral blood mononuclear cells of patients with AA (25 cases) ,PV (12 cases ), and 30 normal individuals. β2-microglobulin (β2M) gene was used as an endogeneous reference. Evaluation of TCR ξ chain gene expression level were used the 2^- △△Ct method. Results: Compared with the normal control, TCR ξ chain gene expression level could be divided into two groups: 10 patients with AA (40%) and 5 patients with PV (41.7%) were over expression, while 15 patients with AA (60%) and 7 patients with PV (58.3%) were down expression. The expression level of TCR chain gene is nonsignificant age-associated in both AA and PV patients. Conclusion:The signaling pathway of T cells activation might display some disorders in patients with AA and PV. The role of the change of TCR ξ chain gene expression pattern in patients with AA and PV should be further confirmed.
