摘要
以不同杂种系的百合品种进行远缘杂交培育杂种。杂种采用Anchored random primer-polymerase chain reaction(ARP-PCR)鉴定,首次报道了利用ARP-PCR技术早期鉴定百合杂种。即利用已经在百合染色体上锚定的引物,扩增亲本和杂种的DNA,鉴定杂种。实验结果表明:ARP-PCR技术中锚定引物的长度是15 bp,退火温度54℃,只扩增出材料的主带型,可重复性高,可靠性好。
Interspecific crosses were made to produce hybrids in Lilium. Anchored random primer-polymerase chain reaction (ARP-PCR) identified the hybrids. DNA of the hybrids and their parents was extracted and amplified with ARP-PCR to identify hybrids. The results showed that ARP-PCR was reproductive and reliable technique for Lilium hybrid identification, which could be the first report of hybrid identification in Lilium with PCR markers . In the ARP-PCR,a single primer of 15bp was used in the PCR reaction,with annealing temperature at 54℃.
