丹参酮Ⅱ A联合顺铂对Hela细胞生长及p53蛋白表达水平的影响 被引量:1

摘要 目的:研究丹参酮Ⅱ A联合顺铂在体外对宫颈癌Hela细胞生长的影响及细胞中p53蛋白表达水平的变化。方法:采用MTT法测定丹参酮Ⅱ A联合顺铂对Hela细胞增殖的影响;HE染色法观察细胞的形态学变化;流式细胞仪检测药物作用后的细胞凋亡情况;Western Blot测定p53蛋白水平的表达。结果:丹参酮ⅡA联合顺铂用药组比同等剂量的两药单用组及对照组对Hela细胞的增殖抑制率高(P<0.05);对照组细胞未见凋亡,而两药单用组及联合组均可见细胞凋亡;药物处理组均可检测到细胞亚G1峰,两药联合组细胞亚G1峰与单药组差异显著;在只含溶媒组、丹参酮Ⅱ A组、顺铂组及丹参酮Ⅱ A联合顺铂组中p53蛋白水平表达逐步升高。结论:丹参酮Ⅱ A联合顺铂比两药单用能显著抑制Hela细胞的生长;二者通过增加p53蛋白的表达,促进细胞凋亡。 Objective To investigate the effect of Tanshinone Ⅱ A (Tan Ⅱ A) combined with Cisplatin (DDP) on the growth and p53 protein expression of Hela cells in vitro. Methods After Hela cells were treated with Tan Ⅱ A, DDP and the combination of Tan Ⅱ A and DDP, respectively, the staining inhibitory effect of them on Hela cells growth was analyzed by MTr assay;the cell apoptosis was measured by HE staining and Flow -cytometry;p53 protein expression in Hela cells was detected by Western Blotting. Results The inhibitory effect on proliferation of Hela cells was significantly enhanced by the combination, HE staining showed the apoptosis induced by Tan Ⅱ A, DDP and the combination, respectively. There was a significant difference in the percentage of apoptotic cells among the Tan Ⅱ A group, DDP group and combination group, Western Blot demonstrated p53 protein expression was increased in the combination group. Conclusion Tan Ⅱ A combined with DDP could enhance the inhibitory effect on proliferation of Hela cells ,the increase protein p53 expression and induction of apoptosis on Hela cells may be the anti- tumor mechanisms of Tan I/A combined with DDP.
