摘要 在硫酸存在下,微量蛋白质的加入可使吡咯红Y—SDS体系弱的共振光散射信号得到极大的增强。在x=364nm处,光散射强度最大,并且光散射增强强度与蛋白质的质量浓度在一定范围内呈线性关系,由此建立了一种测定蛋白质的新的分析方法。对牛血清白蛋白(BSA)和人血清白蛋白(HSA)进行测定,其线性范围分别为0.15-3.6μg/mL和0.06—4.8μg/mL,检测限分别为21.0ng/mL和12.0ng/mL。该方法用于人血清样中蛋白总含量的测定,结果令人满意。 The weak resonance light scattering (RLS) of pyronine Y - SDS could be enhanced greatly by the addition of proteins in the presence of H2SO4, and the maximum scattered wavelengths appears at 364 nm. Based on this, the interaction of pyronine Y - SDS with proteins was studied and a new quantitative determination method for proteins was developed. The enhanced RLS intensity was in proportion to the concentration of proteins in the range of 0.15 -3.6 μg/mL for bovine serum albumin (BSA) and 0.06- 4.8 μg/mL for human serum albumin (HSA) with detection limits of 21.0 and 12.0 ng/mL, respectively. This method was applied to the determination of human serum with good results.
