血竭乳剂对糖尿病小鼠肝脏和肌肉组织中糖原合成酶激酶表达的影响

Effect of Drogon’s Blood on the Expression of GSK-3β in Liver and Muscle of Mice with DM

目的检测糖尿病(Diabetesmellitus,DM)小鼠在接受中药血竭乳剂治疗后肝脏及右后肢肌肉组织匀浆中糖原含量及糖原合成酶激酶(GSK-3β)的表达。方法将120只昆明小鼠分为6组,A组为正常对照组,B组为DM模型组,C、D、E组分别为血竭乳剂低、中、高剂量用药组,F组为盐酸二甲双胍用药组。C、D、E组小鼠分别按125、250、500mg/(kg·d)剂量血竭乳剂灌胃,B组用等体积生理盐水灌胃,F组小鼠按100mg/(kg·d)溶于等体积生理盐水灌胃,1次/d,连续给药7d。结果在给予血竭乳剂后,组织中GSK-3β蛋白表达比DM模型组下降(P<0.05)。结论血竭乳剂能增加DM小鼠肝、肌糖原含量,降低GSK-3β表达水平。

Objective To study the effect of Dragon＇s blood on the expression of GSK-3β and the volume of hepatic glycogen and muscle glycogen in the liver and muscle of mice with DM. Method Selecting 120 mice of Kunming strain, and randomly dividing them into six groups： normal group, DM group, Dragon＇s blood groups with three dosage [125, 250, 500 mg/（kg · d）] and C4H11N5 · HCI group. Dragon＇s blood groups were given Dragon＇s blood by introgastric （ig） everyday for 7 d. C4H11N5 · HCI group were given C4H11N5 · HCI 1 mg/（kg · d） by introgastric （ig） everyday for 7 d. The indices were observed and recorded. Result After given Dragon＇s blood, the expression of GSK-3β was decreased （P〈0.05）. Conclusion Dragon＇sblood could increase the volume of hepatic glycogen and muscle glycogen of mice with DM, inhibit the expression of GSK-3β.
