猪心苹果酸脱氢酶的分离纯化及性质 被引量:2

摘要 采用组织破碎,二度硫酸铵盐析,DEAE Sepharose F.F.离子交换层析及Sephadex G—75 Fine凝胶过滤等纯化方法,从猪心中提取得到了苹果酸脱氢酶。提取纯化的苹果酸脱氢酶经SDS—PAGE测定显示为单一条带,相对分子质量为34000。酶活回收率为57.99%,最适pH为8.0,最适温度为50℃。 By tissue crushing, ammonium sulfate duel-precipitation and purification through DEAE Sepharose F. F. Sephadex G-75 filtration, the malate dehydrogenase (MDH) from swine heart was prepared. The purified MDH was illustrated as a single band in SDS--PAGE. molecular weight of MDH subunit was 34 000.
