摘要 目的:研究激活淋巴细胞和未激活淋巴细胞的端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)的表达情况。方法:运用定量RT-PCR法检测分别代表激活淋巴细胞和未激活淋巴细胞的43枚良性结直肠疾病的炎性系膜淋巴结和31例正常自愿者的外周血单核细胞hTERT的表达水平。另外,作为阳性和阴性对照,还检测了59对结直肠癌肿瘤组织/正常黏膜组织的hTERT表达水平。结果:炎性系膜淋巴结的hTERT表达水平比正常人外周血单核细胞的高,但没有统计学意义(P=0.07)。肿瘤组织的hTERT表达水平明显比正常组织的高(P=0.001)。结论:淋巴细胞无论激活与否,hTERT表达水平没有显著改变,与正常组织也无明显差异。 Objective: To investigate the expression of human telomerase reverse transcriptase (hTERT) in activated lymphocytes. Methods: To investigate the relationship of hTERT expression in activated lymphocytes and non-activated lymphocytes, hTERT mRNA was measured in 43 inflammatory mesenteric lymph nodes from patients with benign large bowel disease and in peripheral mononuclear cell samples from 31 normal donors. hTERT mRNA was also measured in 59 pairs of tumor and normal tissue from colorectal cancer patients as positive and negative control respectively. Results: The hTERT expression in the lymph node group was higher than that in peripheral mononuclear cell group. This difference was not statistically significant (P=0.07). The hTERT expression in tumor tissue was higher than that of matched normal tissue (P=0.001). Conclusion: Higher expression of hTERT may be detected in activated lymphocytes, which was not a significant over-expression when compared to non-activated lymphocytes and normal tissue.
