迟缓爱德华菌eha基因对大肠埃希氏菌溶血基因clyA转录的调控 被引量:2

摘要 目的研究迟缓爱德华菌溶血活化基因eha是否能影响大肠埃希氏菌DH5α溶血素基因clyA的转录。方法将重组eha表达质粒pED101转入DH5α中,以载体质粒pACYC184为对照,观察其溶血情况。并用RT-PCR分别检测受体菌DH5α,含pED101的DH5α菌以及含载体的DH5α菌中clyA基因的转录情况。结果eha基因表达质粒的转入,可以使DH5α出现clyA基因阳性条带,且和载体无关。结论迟缓爱德华菌eha基因能正性调控DH5αclyA基因的转录,从而使其表现为溶血。 Explore whether the hemolysin activator (eha)gene of Edwardsiella tarda can effect the transcription of hemolysin (clyA)gene of Escherichia coli DH5α strain. The hemolytic phenomenon was observed, after the plasmid was transformed into DH5α strain, and then compared with the carrier of plasmid pACYC184. The transcription of the hemolysin gene (clyA)from receptor transformed with eha gene was tested by RT-PCR, so did the transcription of clyA from the receptor with carrier pACYC184 and from the receptor with no carrier. The results showed that the positive strap of clyA gene was only seen in the receptor strain transformed with eha gene. In dicating that the eha gene of Edwardsiella tarda can regulate positively the transcription of clyA gene so that its carrier was shown hemolytic.
