摘要
目的建立外源性曲霉菌性眼内炎的豚鼠动物模型。方法选取健康成年的豚鼠(体重400~450g)28只,随即分为7组,右眼为实验眼,分别于玻璃体腔内接种浓度为(1.0~1.5)×103CFU.mL-1、(10~15)×103CFU.mL-1、(100~150)×103CFU.mL-1、(1.0~1.5)×106CFU.mL-1、(10~15)×106CFU.mL-1、(100~150)×106CFU.mL-1、(1.0~1.5)×109CFU.mL-1的孢子悬液各0.02mL,左眼为正常对照眼,注射生理盐水0.02mL。采用直接检眼镜进行自然病程及临床特征观察15d,处死后取玻璃体腔液、全身主要脏器及血液行真菌培养,进行真菌形态学及分子生物学鉴别,并将眼球做组织学检查。选取临床特征典型且无全身感染的实验组浓度进行重复试验。结果(1.0~1.5)×106CFU.mL-1组眼部组织的感染症状典型,主要内脏和血液培养阴性,眼部组织浓汁培养出病原菌阳性率100%.结论成功建立了外源性曲霉菌性眼内炎的豚鼠模型。
Objective To establish exogenous aspergillus fumigatus endophthalmitis model of guinea-pig. Methods Twenty-eight healthy guinea-pigs, weight of 400 - 450 g, were randomly divided into seven groups according to the aspergillus fumigatus albicans suspension concentration of ( 1.0 - 1.5) × 10^3 CFU · mL^-1 ,(10 - 15) × 10^3 CFU · mL^-1 , ( 100 - 150) × 10^3 CFU · mL^-1 , ( 1.0 - 1.5)× 10^6 CFU · mL^-1 ,( 10 - 15) × 10^6CFU · mL^-1 ,( 100 - 150) × 10^6CFU· mL^-1, ( 1.0 - 1.5) × 10^9 CFU· mL^-1(colony forming unit, CFU). Normal saline 0.02 mL was injected into left eyes as control groups and 0.02 mL aspergillus fumigatus albicans suspension was injected into right eyes as the experimental groups respectively. The course of disease and clinical feature had been observed by direct ophthalmoseope for 15 days. AU guinea-pigs were executed to make fungi culture of vitreous,blood and main organs. And the cultured fungi were identified by morphology and molecttlar biology. Histopathologies of the eyes were examined. Repeated experiment was taken in the concentration of experimental groups with typical clinical features but without systemic infection. Results Typical fungal infectious symptoms of eye without blood infection were observed in the group of the concentration of ( 1.0 - 1.5 ) × 10^6 CFU · mL^-1 and the positive rate of vitreous culture was 100%. Conclusion Exogenous aspergillus fumigatus endophthalmitis model in a guinea-pig is successfully established.
