摘要
目的通过观察血必净对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)患者T淋巴细胞增殖及分泌白细胞介素-2(IL-2)、表达CD25的影响,为血必净对COPD的治疗提供理论依据。方法采用酶联免疫吸附试验法测定血必净对T淋巴细胞分泌的细胞因子IL-2影响,采用流式细胞仪测定血必净对T淋巴细胞表达CD25影响,用四甲基偶氨唑盐(MTT)比色法观察血必净对T淋巴细胞增殖的影响。结果COPD患者T淋巴细胞生成的IL-2、表达CD25水平与对照组比较显著减低(P<0.01),T淋巴细胞增殖反应亦明显减低(P<0.01);血必净和慢阻肺组,PHA刺激T淋巴细胞,T淋巴细胞生成IL-2、表达CD25的较慢阻肺组高(P<0.01),T淋巴细胞增殖反应亦明显增加(P<0.01)。结论COPD患者T淋巴细胞生成的IL-2、表达CD25减低,T淋巴细胞增殖反应亦减低,免疫失衡;血必净促进COPD患者T淋巴细胞分泌IL-2、表达CD25,增强T淋巴细胞增殖反应。
Objective Toobserve the effect of xuebijing on T lymphocyte function in patiens with acute exacerbation chronic obstructive pulmonary diseases (COPD). Method IL- 2 of T lymphocytes in patients with COPD were determined with the method of enzyme- linked immunoadsordent assay kits (ELISA) ; the activation markers CD25 were analyzed by flow cytomery; proliferation assays of T lymphocytes were determined by MTT assay. Results In COPD patients , IL - 2 levels, the activation markers CD25, the proliferation assays were significantly lower than those of normal control group ( P 〈0.01). We added xuebijing to PHA cultures ofT cells from COPD patients , IL- 2 levels, the activation markers CD25 andthe proliferation assays T lymphocytes have increased in cultures, compared to PHA cultures of T cells from COPD group. Conclusions IL - 2 levels, the activation markers CD25 and the proliferation assays are abnormal in COPD patients, xuebijing can enhance IL - 2 levels, the activation markers CD25 and the proliferation assays , so improve T lymphocytes function in COPD patients.
