bFGF对小鼠ES细胞生长增殖的影响 被引量:1

摘要 【目的】研究碱性成纤维细胞生长因子(bFGF)对ES细胞增殖的影响,为建立稳定的小鼠ES细胞培养和扩增打下基础。【方法】将E14小鼠胚胎干细胞分别培养于含LIF10^6U/L并含5μg/L、10μg/L、20μg/L 3种浓度的bFGF的ES细胞基础培养液中,观察细胞形态和计数ES细胞克隆数。【结果】10μg/L和20μg/L bFGF与含LIF 10^6U/L的ES细胞形态相似,3d后ES细胞克隆数分别为:88±8、128±12和114±17,三者克隆数无显著差异(P〉0.05)。ES细胞组化染色呈AKP和SSEA-1阳性。【结论】bFGF可促进小鼠ES细胞增殖,其最佳浓度为10μg/L。
