树突状细胞体外诱导抗前列腺癌免疫的研究

摘要 【目的】研究树突状细胞(DC)在体外诱导抗前列腺癌的免疫作用。【方法】自前列腺癌患者外周血中分离 CD34+干细胞,加入细胞因子(rhGM-CSF,rhTNF-α及 rhlL-4)培养;以人前列腺癌细胞系 LNCap 细胞的肿瘤相关抗原(TAA)激活 DC;DC 诱导自体 T 淋巴细胞增殖、分化为细胞毒素性 T 细胞(CTL);检测 CTL 对 LNCap 细胞、HepG2细胞、LOVO 细胞及HOS-8603细胞的细胞毒作用。【结果】前列腺癌患者外周血 DC 能够诱导自体 T 淋巴细胞增殖分化为 CTL,该 CTL 对 LNCap细胞有强大的杀伤力(杀伤率为89%±10%,),对 HepG2细胞,LOVO 细胞及 HOS-8603细胞则无明显的细胞毒作用(杀伤率分别为10%±3%,8%±6%,6%±4%)。【结论】前列腺癌患者外周血 DC 体外能够诱导高效而特异抗前列腺癌免疫。提示DC 可能在治疗前列腺癌及预防前列腺癌术后复发和转移中发挥重要作用。
