摘要
目的探讨通心络胶囊抑制炎性因子表达以防治动脉粥样硬化(AS)的作用。方法球囊拉伤加高脂饮食12周建立兔AS模型,通心络胶囊组、辛伐他汀组分别予相应的药物干预,对照组仅予普通饲料,共24周。采用酶联免疫吸附(ELISA)法检测血清中血管间细胞黏附分子-1(VCAM-1)、细胞间细胞黏附分子-1(ICAM-1)和超敏感C反应蛋白(hs-CRP)的浓度,免疫组化方法检测斑块内VCAM-1和ICAM-1的表达情况。结果血清VCAM-1、ICAM-1和hs-CRP的浓度较用药前均显著降低,对照组前后无变化;两药物组与对照组比较,各项检测内容均有显著差异。免疫组化结果与其相似。免疫组化显示,对照组斑块ICAM-1和VCAM-1内呈强阳性表达,药物组表达较弱。结论通心络胶囊能有效抑制炎症反应,与他汀类药物相比无显著差异。
Objective To investigate the effects of "Tongxinluo Capsule" (TXL) on prevention and treatment of atherosclerosis (AS) by inhibiting expression of inflammatory factors. Methods Rabbits' atherosclerosis plaques model was made by balloon-induced abdominal aorta wall injury and a diet of high cholesterol for 12 weeks, TXL group and simvastatin group received corresponding drugs, control group was given normal food and the trial lasted 24 weeks. Adopt ELISA kit to detect the concentration of vascular cellular adhesion molecule 1 (VCAM-1), intercellular cellular adhesion molecule 1 (ICAM-1) and high sensitivity C reaction protein (hs-CRP) in blood serum, discover the expressing information of VCAM-1 and ICAM-1 in plaque by immunohistochemistry. Results After medication, the concentration of VCAM-1, ICAM-1 and hs-CRP was reduced, but there was no significant difference in the control group; compared with the two drug treatment groups with control group, all detection contents were markedly different. The results of immunohistochemistry were similar: the ICAM-1 and VCAM-1 expressed strongly in the control group and weakly in two drug groups. Conclusion "Tongxinluo Capsule" can inhibit the inflammatory reaction as effectively as statins do.
