骨髓干细胞移植治疗心肌梗死后心衰的实验研究 被引量:3

摘要 目的探讨骨髓干细胞移植治疗心肌梗死后大鼠心力衰竭的效果。方法取SD雄性幼鼠10只作为骨髓干细胞的供体。经过骨髓干细胞的分离、培养和标记制成每毫升含10×108的细胞悬液用于移植。将SD雄性大鼠60只作为细胞移植的受体,随机分成假手术组、心肌梗死组、细胞移植组,每组20只。心肌梗死组、细胞移植组大鼠建立心肌梗死模型,假手术组未结扎冠状动脉。细胞移植组吸取0.2mL骨髓干细胞悬液注射到瘢痕组织中,心肌梗死组注入等量干细胞培养液基质,假手术组不予任何移植处理。术后4周,采用超声心动图和解剖直测法获得心梗组和移植组大鼠心脏功能、心室重构和病理学资料。结果超声图和解剖直测值的结果均显示,与假手术组相比,心梗组大鼠心室腔扩大,室壁变薄,球形化。超声结果显示,心梗大鼠心脏功能明显降低。与心梗组相比,移植组大鼠心室重构程度减低,心功能有所改善。结论骨髓干细胞移植能改善心梗后的心室重构和心功能。 Objective In order to illustrate whether bone marrow stem cells(BMSC) could treat heart failure in rats with myocardial infarction(MI). Methods Ten SD young rats weighting (50 ± 10)g were selected as doner for cellular transplantation. BMSC was cultured and labeled ex vivo. BMSC pellet was resuspended in culture medium at a concentration of 10 × 10^8 BMSCs in lmL for cell transplantation. Sixty SD rats weighting (300 ± 50)g were selected as the receptor of cell transplatation and randomly divided into 3 groups with 20 rats in each group: The sham operating group, MI group and transplant group. MI group and transplanting group rats were operated and left coronary artery were ligated. Three weeks after the operation, either 0.2mL cell suspen- sion (transplant group) or culture medium(MI group) was injected into the scared tissue by Using lmL tuberculin syringe, the sham operating group underwent chest opening but no ligating coronary artery or transplantation with cells or culture medium. Four weeks after the cell transplantation, the heart function and ventricular remodeling and pathologies results in all groups were measured by ultrasonic cardiography(UCG) and direct survey. Results The both of ultrasonic cardiography(UCG) and direct survey Tesults showed left ventricular (LV) chambers were more extended and LV free walls became thinner and shape of LV became more obviously spherical and heart functions were more decreased in MI group than the sham operating group. LV was less remodehng and LV functions were better in transplant group than in MI group. Conclusion The bone marrow stem cell transplantation could improve the LV remodeling and heart function after myocardial infarction.
