摘要
以高氏白澳山龙眼Leucadendron galpinii幼苗为外植体,采用正交设计试验,研究不同因子的不同水平对高氏白澳山龙眼丛芽增殖与生根的影响。结果表明,外源激素中,6-BA对丛芽分化影响最大,在0.5-1.0mg·L^-1,其丛芽分化随6-BA浓度的升高而增多,而在1.0-2.0mg·L^-1则反之;NAA对丛芽分化影响不明显;蔗糖、抗坏血酸均随浓度升高丛芽分化增多。经方差分析显示,最佳丛芽增殖配方为:1/2MS+6-BA1.0mg·L^-1+NAA0.1mg·L^-1+蔗糖30g·L^-1+抗坏血酸15mg·L^-1。IBA对生根培养影响最大。在光照培养条件下,最佳生根培养配方为:IBA1.5mg·L^-1+改良MS+蔗糖30g·L^-1。
The seedlings of Leucadendron galpinii were used as the explants and the effects of different levels of the factors on axillary shoot proliferation and rooting.were studied with orthogonal design. The results showed that the effect of 6 -BA was the biggest among the external hormones. The axillary shoot proliferation increased with the increasing of 6- BA concentrations in the range of 0.5-1.0mg·L^-1 and was contrary in the range of 1.0 -2.0mg·L^-1. The effect of NAA was not significant on axillary shoot proliferation. As the concentrations of sucrose and ascorbic acid were increasing,the axillary shoot proliferation increased significantly. According to the results of variance analysis, the optimal medium for axillary shoot proliferation showed as follows : 1/2MS + 6 - BA 1.0mg·L^-1 + NAA 0.1mg·L^-1 + sucrose 30g·L^-1 + ascorbic acid 15mg·L^-1. The effect of IBA on rooting was the biggest and the optimal medium for rooting was IBA 1.5mg·L^-1 + modified MS + sucrose 30g·L^-1,under the condition of light.
