摘要
目的探讨胃癌患者体内CD4+CD25+调节性T细胞的含量及FoxP3 mRNA在Treg中的表达情况及其临床意义。方法应用流式细胞术(FCM)测定20例胃癌患者外周血中Treg细胞含量,同时采用RT-PCR技术检测其FoxP3 mRNA的表达情况,并与20例健康志愿者的外周血进行对比研究和统计学分析。结果胃癌患者外周血中Treg细胞占CD4+T细胞总数的(12.76±0.46)%,显著高于健康对照组的(6.81±0.66)%,(P<0.01);FoxP3 mRNA在胃癌患者外周血Treg细胞中呈现高表达(0.84±0.02)%,明显高于其在健康对照组中表达(0.76±0.03)%,(P<0.05)。结论胃癌患者外周血中Treg的数量较正常人明显增高,同时伴随有FoxP3 mRNA高表达,提示FoxP3基因可能对Treg有重要的调节功能,从而影响胃癌病人的抗肿瘤自身免疫功能。
Objective To investigate the contents of CD4+ CD25+ T cells and the expression of FoxP3 mRNA in these Treg cells in peripheral blood of the patients with gastric cancer and to characterize their clinical significance. Methods By using flow cytometry, the levels of CD4+ CD25+ T cells in peripheral blood of 20 preoperative patients with gastric cancer were determined. RT-PCR techniques were used to detect the expression of FoxP3 mRNA in these Treg cells, and the results were analyzed statistically in comparison with that of 20 healthy volunteers. Results The percentage of CD4+ CD25 + T cells in total CD4+ T cells isolated from the patients with gastric cancer was (12. 76 ± 0. 46) %, in comparison, it was (6. 81 ± 0. 66) % in those from healthy volunteers. It was obviously different between Treg cells amount of gastric patients and that of volunteers (P〈0. 01). The FoxP3 mRNA expression of Treg cells in the patients with gastric cancer (0. 84 ± 0. 02)%] was significantly higher than that in healthy volunteers [(0. 76± 0. 03) %, (P〈0. 05). Conclusion The levels of CD4+ CD25+ T cells in peripheral blood and the FoxP3 mRNA expression of Treg cells in gastric cancer patients were increasing significantly. FoxP3 genes may play a pivotal role in regulating the function of Treg cells in self-tumor immunity of the patients with gastric cancer.
