摘要
研究了影响七种核酸碱基-胸腺嘧啶,尿嘧啶,胞嘧啶,腺嘌呤,鸟嘌呤,黄嘌呤和次黄嘌呤毛细管电泳分离的因素。在24.5℃下,七种碱基在40mmol/L十二烷基硫酸钠-10mmol/L硼砂-17%甲醇-3.0mmol/Lβ-环糊精(pH9.30)缓冲液中电泳可得良好分离。保留时间的变异系数小于1.8%(n=6)
The retention behaviours of seven common bases thymine,uracil,cytosine,adenine,guanine,xanthine and hypoxanthine were investigated by micellar electrokinetic capillary chromatography.At 24.5℃ and 20kV applied voltage,7 bases introduced into the separation carillary (65cm×50 μm i.d.,51.5cm to UV detector)were separated satisfactorily within 21 minutes in 40 mmol/L SDS 10mol/L borax 17% methanol 3 0mmol/L β cyclodextrin buffer solution (pH9.30).The detection limits of the bases were within 0.6~6.0mg/L,and the relative standand deviation of retention times and UV absorbances of the bases were less than 1.8% and 6.5% respectively.
