摘要
以微型月季品种‘白珂斯特’(‘White Koster’)、‘金太阳’(‘Golden Sun’)和‘冬梅’(‘Winter Plum’)为试材,采用无机酸法、TTC法、培养法、碘-碘化钾法、形态观察法和混合"活染"法测定其花粉生活力。结果表明:(1)上述6种方法测得的‘白珂斯特’花粉生活力分别为0%、0%、7.98%、38.16%、60.48%和77.40%,‘金太阳’花粉生活力的结果分别为0%、0%、10.67%、22.92%、46.59%和73.28%,‘冬梅’花粉生活力分别为0%、0%、14.18%、18.79%、69.89%和80.35%,表明不同方法差异很大。(2)混合"活染"法和形态观察法适用于微型月季花粉生活力的检测,碘-碘化钾法、无机酸法和TTC法不适用于微型月季花粉生活力的检测;培养法虽然是测定花粉生活力的理想方法,但本实验采用的培养基(15%蔗糖,0.01%硼酸,10%的琼脂)还不是微型月季花粉生活力的最佳培养基有待进一步优化。
Six measurement methods are used to determine pollen viability of three cultivars of minirose (‘ White Koster',‘ Golden Sun' and ‘Winter Plum'). These methods are Inorganic acid method, TTC staining, in vitro culture, I-KI staining, morphological observation and blend staining. The results of the research indicate that: (1)There are great differences among different measurement methods and different cultivars of minirose. As to ‘White Koster', the pollen viability are 0%, 0%, 7.98%, 38.16%, 60.48%, and 77.40% respectively; As to ‘Golden Sun', the pollen viability of six methods are 0%, 0%, 10.67%, 22.92%, 46.59%, and 73.28%;As to Winter Plum, the pollen viability of six methods are 0%, 0%, 14.18%, 18.79%, 69.89%, and 80.35% respectively. (2)Blend staining and Morphological observation are suitable for detecting pollen viability of minirose. Methylene blue staining,inorganic acid method and Trc staining are not suitable for detecting pollen viability of Rosa chinensis var. minima. Although in vitro culture is an ideal method for detecting pollen viability of plant species, the media used in this study were not the optimal ones and need to be further optimized.
