摘要 建立了高效毛细管电泳法测定侧柏中槲皮素含量的方法,确定了槲皮素的最佳提取方法,即微波强化萃取法.通过研究缓冲溶液酸度、浓度、工作电压和进样时间等对毛细管电泳分离效率的影响,优化了分析条件.以30mmol/L的硼酸-硼砂缓冲溶液(pH=8.62)为电泳介质,进样压力为0.5psi,进样时间8s,在25℃和25kV恒压下进行电泳分离,于254nm波长处检测.在此条件下,7min内实现了侧柏中槲皮素的良好分离.槲皮素峰面积和质量浓度在4.0-300mg/L范围内成良好线性关系,检出限为0.5mg/L.方法简单、快速、准确、线性范围宽,用于侧柏中槲皮素的测定,结果令人满意. In this paper, the author developed a method for the determination of quercetin in arborvitae by the high performance capillary electrophoresis and the best way to extract quercetin, namely, the microwave assistance extracting technique. The effect of buffer aqua acidity and concentration, applied voltage and injection time were investigated. With the buffer aqua of the boric acid and borax that implied a 30mmol/L(pH 8.62) as electrophoresis medium, quercetin was separated with a temperature of 25 ~C and a constant pressure of 25kV. Then it was examined in the 254nm wavelength. Under the optimized conditions, good separation was found in less than 7 minutes.A good linearity between peak current and concentration was found in the 4.0-300mg/L. The detection limit was 0.5mg/L for quercetin. The method is simple, fast, accurate, line scope breadth, and has been applied to the determination of amount of the quercetin in arborvitae samples with satisfactory results.
