摘要
目的:通过观察不同浓度的氢化可的松对人A375黑素瘤细胞的生长、黑素生成以及酪氨酸酶活性的影响,探讨氢化可的松治疗白癜风的作用机制。方法:用不同浓度的氢化可的松处理人A375黑素瘤细胞,通过MTT比色法观察氢化可的松对人A375黑素瘤细胞生长的影响,用NaOH裂解法测定黑素的含量,用多巴氧化法测定酪氨酸酶活性的变化。结果:氢化可的松在一定浓度范围内促进人A375黑素瘤细胞增殖,酪氨酸酶活性增强,黑素含量增加。结论:一定浓度的氢化可的松可以促进人A375黑素瘤细胞增殖,激活酪氨酸酶活性,促进黑素生成,为探讨氢化可的松治疗白癜风的机制提供了实验依据。
Objective To observe the effects of hydrocortisone in different concentration on the cell growth, melanogenesis and tyrosinase activity in A375 human melanoma cells for dissecting the mechanism of hydrocortisone in treating vitiligo. Methods After the A375 melanoma cells were dealed with hydrocortisone in different concentrations, we observed the growth of A375 melanoma cells by MTT assay; measured the contents of melanin by NaOH lysis method; and detected the change of tyrosinase activity by dioxyphenylalanine oxidation. Results Compared with the control group, hydrocortisone concentration in a certain extent enhanced the proliferation of A375 human melanoma cells, tyrosinase activity and melanogenesis. Conclusion Hydrocortisone of certain concentration car~ promote the proliferation of A375 human melanoma cells and activate the activity of tyrosinase, enhence the melanogenesis, and the experiment has provided an experimental basis for exploring the mechanism of hydrocortisone in treating vitiligo.
