Rubratoxin B诱导人绒癌细胞株凋亡及Bcl-2 mRNA表达的研究

摘要 目的:定量分析rubratoxin B(RB)在体外诱导人绒癌细胞株(BeWo)发生凋亡,并探讨其与抑制凋亡基因Bcl-2mRNA表达的关系。方法:利用MTT、HE染色、annexinV-FITC/PI流式细胞技术定性及定量分析经RB处理后BeWo细胞活细胞及凋亡细胞的变化情况;利用逆转录聚合酶链式反应(RT-PCR)技术检测抑制凋亡基因Bcl-2与RB诱导的BeWo细胞凋亡的关系。结果:RB诱导BeWo细胞凋亡的数量与药物浓度及用药时间明显相关。RT-PCR结果显示Bcl-2mRNA的表达随着凋亡细胞的增加而减少。结论:RB能够诱导BeWo细胞发生凋亡,Bcl-2mRNA表达的下调是RB诱导BeWo细胞凋亡的重要机制。 Objective To quantitatively analyze rubratoxin B (RB) - induced apoptosis in human choriocarcinoma cell line BeWo in vitro and to explore its relation with Bcl-2 mRNA expression. Methods The BeWo cells were qualitatively and quantitatively analyzed by MTT assay, HE staining, and annexinV-FITC/PI-FCA technique after being treated with RB. We used RT-PCR to determine the relation between Bcl-2 mRNA expression and apoptosis of BeWo induced by RB. Results The amount of the apoptotic cells was markedly related with RB in a doseand time-dependent manner. RT-PCR revealed a decline in Bcl-2 mRNA expression with the increasing apoptosis of the cells. Conclusion Rubratoxin B can induce apoptosis of BeWo cells, whose mechanism involves the downregulafion of Bcl-2 mRNA.
