摘要
通过单因素和正交试验对芽孢杆菌B50产木聚糖酶的培养基配方和发酵条件进行研究。结果表明:5%麸皮为最佳碳源,1%(NH4)2HPO4为最佳氮源,酵母浸粉0.2%,NaCl0.6%,K2HPO40.2%,CaCl20.01%,FeSO40.01%,MgSO40.01%,初始pH7.0,接种量为5%,100mL三角瓶装液量为20mL;最佳培养温度为25℃,培养时间60h。其木聚糖酶活力可达到15362.6U/mL。
The fermentation conditions of xylanase by Bacillus sp. B50 are optimized in the study through the single factor and the orthogonal experiment. So the best conditions are: carbon source: 5% bran, nitrogen source: 1% (NH4)2HPO4, yeast 0.2%, NaCI 0.6%, K2HPO4 0.2%, CaCI2 0.01%, FeSO4 0.01%, MgSO4 0.01%, start pH7.0, the inoculation content 5%, 20mL in 100mL triangular flask, the best temperature is 25℃ and the time is 60h. So the xylanase activity can reach about 15362.6U/mL.
