高效液相色谱法测定苦荞籽壳中芦丁和槲皮素的含量

HPLC determination of rutin and quercetin in buckwheat husks

目的:建立高效液相色谱法测定苦荞籽壳中芦丁和槲皮素的含量。方法:采用 Zorbax SB-C_(18)(4.6 mm×150 mm,5μm)色谱柱;流动相为甲醇-0.4%磷酸(50:50);流速1.0 mL·min^(-1);检测波长254 nm。结果:芦丁进样量在0.024～0.963mg·mL^(-1)范围内(r=0.9996),槲皮素进样量在0.0132～0.528 mg·mL^(-1)范围内(r=0.9990)线性关系良好,平均回收率芦丁为100.1%(RSD=1.15%),槲皮素为99.9%(RSD=1.04%)。结论:该方法操作简便,分离度高,重复性好,可同时测定苦荞籽壳中芦丁和槲皮素的含量。

Objective：To develop an HPLC method for determination of rutin and quercetin in buckwheat husks. Methods：A Zorbax SB - C18column （4. 6 mm × 150 mm,5 μm） was used with a mobile phase of methanol - 0. 4% phosphoric acid solution （50： 50） ,the flow rate was 1.0 mL . min^-1 ,the UV detector was set at 254 nm. Results： A linear relationship of rutin was good at the range of 0. 024 - 0. 963 mg . mL^ - 1 （ r = 0. 9996 ） and quercetin was＇good at the range of 0. 0132 -0. 528 mg . mL^-1（ r = 0. 9990）. The average recovery of rutin was 100. 1% （RSD = 1.15% ） and of quercetin was 99.9% （RSD = 1.04% ）. Conclusion：The method is simple, accurate and good reproducibility. It is suitable for the simultaneous determination of rutin and quercetin in buckwheat husks.
