大肠癌中P53、nm23-H1和MMP-9基因的表达及其临床意义

Expression of P53,nm23-H1 and MMP-9 in colorectal adenocarcinoma and its clinical significance

目的研究P53、nm23-H1和MMP-9基因在大肠癌中的表达及其与患者的临床病理和预后的关系。方法采用免疫组化LSAB法检测78例大肠癌标本及30例正常粘膜的P53、nm23-H1和MMP-9的基因蛋白表达。结果(1)P53、nm23-H1、MMP-9的阳性表达率在大肠癌中分别为56.41%、39.74%和58.97%,在正常肠粘膜中的表达率分别为13.33%、83.33%和6.67%,其差别有显著性意义(P<0.01)。(2)P53和MMP-9的表达与肿瘤的浸润深度和淋巴结转移呈正相关(P<0.01),nm23-H1的表达与肿瘤的浸润深度和淋巴结转移呈负相关(P<0.01)。结论联合检测大肠癌中P53、nm23-H1和MMP-9基因表达,对了解大肠癌的生物学特性和预后判断有重要意义。

Objective To study the expression and significance of P53 ,nm23-H1 and MMP-9 in colorectal carcinoma and their correlations with clinical pathologic parameters and prognosis. Methods Expressions of p53, nm23-H1 and MMP-9 were detected in 78 cases of colorectal adenocarcinoma and in 30 cases of normal mucosa by immunohistochemical staining （LSAB method）, Results （1） The positive rates of P53 and MMP-9 expression in the colorectal adenocarcinoma （56.41% and 58.97% respectively） were significantly higher than those in the normal tissues（ 13.33% and 6.67% respectively） （P 〈 0.01 ）. The positive rate of nm23-H1 （39.74%） expression in the colorectal adenocarcinoma was significantly lower than that in the normal tissues （83.33%） （ P 〈 0.01 ）. （2） The positive rates of P53 and MMP-9 expression were positively correlated with metastasis of lymph nodes and adenocarcinoma infiltration （ P 〈 0. 01 ）. The positive rate of nm23-H1 expression was negatively correlated with metastasis of lymph nodes and adenocarcinoma infiltration（P 〈0.01 ）. Conclusion The combined detection of P53, nm23-H1 and MMP-9 in colorectal adenocarcinoma may be helpful for the understanding of biological characteristics of colorectal adenocarcinoma and prognosis of patients.
